Penicillin production by high-yielding strains of Penicillium chrysogenum.

چکیده
The development and study of strains of Penicillium chrysogenum that give high yields of penicillin has been a major project at the University of Wisconsin during the past 12 years. Many thousands of cultures have been screened for their penicillin-producing ability in the Botany Department of the University. The methods used for the production and screening of mutants, the genealogy of the outstanding strains and their cultural characteristics have been described recently by Backus and Stauffer (1955). In recent years, interest has centered on cultures that produce no pigment but give high yields of penicillin. Many of the superior cultures have been studied intensively both in shaken flasks and pilot plant fermentations in the Biochemistry Department (Anderson et al., 1953, 1956; Davey and Johnson, 1953; Soltero and Johnson, 1954; and Owen and Johnson, 1955). In the present paper we shall report on factors affecting penicillin production by some of the recent high-yielding strains. These are numbered W50-935, W50-1583, W51-20, W51-616, W51-20F3, and W51-20F3-64.

Increased penicillin production in Penicillium chrysogenum production strains via balanced overexpression of isopenicillin N acyltransferase.
Intense classical strain improvement has yielded industrial Penicillium chrysogenum strains that produce high titers of penicillin. These strains contain multiple copies of the penicillin biosynthesis cluster encoding the three key enzymes: δ-(l-α-aminoadipyl)-L-cysteinyl-D-valine synthetase (ACVS), isopenicillin N synthase (IPNS), and isopenicillin N acyltransferase (IAT). The phenylacetic aci...

متن کاملNonlinear biosynthetic gene cluster dose effect on penicillin production by Penicillium chrysogenum.
Industrial penicillin production levels by the filamentous fungus Penicillium chrysogenum increased dramatically by classical strain improvement. High-yielding strains contain multiple copies of the penicillin biosynthetic gene cluster that encodes three key enzymes of the β-lactam biosynthetic pathway. We have analyzed the gene cluster dose effect on penicillin production using the high-yieldi...
متن کاملSulfate utilization by penicillin-producing mutants of Penicillium chrysogenum.
A great deal of work has been directed toward increasing the yield of penicillin in fermentations with Penicillium chrysogenum. The spectacular increase in yield which has been achieved has resulted, in the main, from two approaches. In the first approach, mutant strains of P. chrysogenum produced by spontaneous variation or by the action of mutagenic agents such as nitrogen mustard or ultravio...
متن کاملComparative Study of Penicillin Production with Vegetative and Spore Inoculum of Penicillium chrysogenum1
In commercial penicillin production the fermentation medium is usually seeded with a mycelial (vegetative) inoculum of Penicillium chrysogenum prepared by the following transfers: spores -+ germinator tank -seed tank. However, depending on the fermentation conditions, the operation in the germinator tank can be successfully eliminated. Goldschmidt and Koffler (1950) used spore-inoculated fermen...
